Pharmacological Mechanism: Ergosterol Synthesis Inhibition

Clotrimazole works by disrupting the structural integrity of the fungal cell membrane:

• Enzyme Blockade: It inhibits the fungal cytochrome P450 enzyme, 14$\alpha$-demethylase.

• Ergosterol Depletion: This enzyme is responsible for converting lanosterol to ergosterol, an essential component of the fungal cell membrane (equivalent to cholesterol in humans).

• Membrane Permeability: The resulting depletion of ergosterol and accumulation of methylated precursors lead to increased membrane permeability.

• Cell Lysis: The cell begins to leak vital intracellular phosphorus compounds and potassium, leading to the inhibition of growth (fungistatic) or cell death (fungicidal) at higher concentrations.
